What is Amdipine 5mg?

Amdipine 5mg Tablets contain Amlodipine Besylate, a dihydropyridine calcium channel blocker (CCB) used for the management of hypertension (high blood pressure) and stable and variant (Prinzmetal) angina. Amlodipine has a long half-life of 35–50 hours, making once-daily dosing effective with gradual, sustained blood pressure reduction and minimal reflex tachycardia. 5mg is the standard starting dose for most patients. How Does Amdipine Work?

Amlodipine selectively inhibits L-type calcium channels in vascular smooth muscle, reducing calcium entry and causing peripheral arterial vasodilation. This decreases peripheral vascular resistance (afterload), lowers blood pressure, and improves coronary artery blood flow — relieving myocardial ischaemia and anginal symptoms without significantly depressing heart rate or myocardial contractility.

Indicated for: essential hypertension (monotherapy or in combination), chronic stable angina, variant angina. Suitable for elderly patients and those with mild-to-moderate renal impairment. Side Effects

Common: peripheral oedema (ankle swelling — most frequent CCB side effect, dose-dependent), headache, flushing, dizziness, fatigue. Less common: nausea, palpitations, somnolence. Rare: gingival hyperplasia, hepatitis. Oedema is caused by arteriolar dilation (not fluid overload) and may not respond to diuretics; dose reduction or agent switch may help.

Caution in: severe hepatic impairment (use lower dose; half-life prolonged), hypotension, severe aortic stenosis. Generally well-tolerated in elderly and renal impairment. Avoid grapefruit juice (CYP3A4 inhibition raises amlodipine levels). Drug interactions: simvastatin (co-administration with amlodipine limits simvastatin to 20 mg/day), cyclosporine, tacrolimus.
