Description

Profy 15mg Tablets — Aripiprazole (Atypical Antipsychotic)

Profy 15mg Tablets contain Aripiprazole, an atypical antipsychotic with a uniquely differentiated mechanism: it is a dopamine D2/D3 partial agonist rather than a pure antagonist. This partial agonism means it stabilises dopamine signalling — functioning as a functional antagonist in dopamine-overactive pathways (mesolimbic) while providing partial agonism in dopamine-deficient pathways (mesocortical). Additionally, aripiprazole is a 5-HT1A partial agonist and 5-HT2A antagonist, providing mood-stabilising and antidepressant benefits.

Approved Clinical Indications

  • Schizophrenia — acute and maintenance treatment
  • Bipolar I Disorder — acute manic and mixed episodes
  • Adjunct treatment for Major Depressive Disorder (MDD) — added to antidepressant therapy
  • Irritability associated with Autism Spectrum Disorder (ASD)

Advantages vs Other Atypical Antipsychotics

  • Lower metabolic burden: Minimal weight gain and less glucose/lipid dysregulation than olanzapine or quetiapine
  • Less sedation: Activating rather than sedating for most patients
  • Less prolactin elevation: Lower risk of hyperprolactinaemia-related side effects
  • No significant QTc prolongation at therapeutic doses

Important Clinical Notes

  • Akathisia (restlessness): The most common and potentially dose-limiting side effect — may require dose reduction or adjunct treatment (beta-blocker or benzodiazepine). Distinguish from agitation.
  • Nausea and insomnia: Common early in treatment, usually transient.
  • Impulse control disorders (rare): Pathological gambling, hypersexuality — discontinue and review if these emerge.
  • CYP2D6 and CYP3A4 interactions: Aripiprazole dose adjustment required with strong inhibitors (fluoxetine, paroxetine) or inducers (carbamazepine).

Usage & Benefits
  • Unique dopamine stabilisation: D2/D3 partial agonism — acts as functional antagonist in overactive pathways while supporting dopamine function in deficient pathways
  • Lower metabolic risk: Significantly less weight gain, glucose dysregulation, and dyslipidaemia than olanzapine or quetiapine
  • Reduced sedation: Activating profile — less daytime somnolence, suitable for patients who cannot tolerate heavy sedation
  • Lower prolactin elevation: Fewer hyperprolactinaemia-related side effects (galactorrhoea, sexual dysfunction)
  • Broad indication: Schizophrenia, bipolar mania, adjunct MDD, and ASD irritability
How To Use

Take exactly as prescribed by your psychiatrist — typically once daily with or without food. Swallow the tablet whole with water. Do not stop or adjust dose without medical guidance. Akathisia (restlessness) is a common early side effect — inform your prescriber if this occurs, as dose adjustment or adjunct treatment may be needed. Monitor for impulse control changes (rare but important). Aripiprazole interacts with CYP2D6 and CYP3A4 inhibitors and inducers — review all co-medications with your prescriber.

FAQs

How is Aripiprazole different from other antipsychotics?

Aripiprazole is a dopamine D2/D3 partial agonist — it stabilises dopamine activity rather than fully blocking it. This explains its lower risk of metabolic side effects, less sedation, and less prolactin elevation compared to full D2 antagonists like olanzapine or quetiapine.

What is akathisia and how common is it with Aripiprazole?

Akathisia is a feeling of restlessness and inability to sit still. It is the most common dose-limiting side effect of aripiprazole and may require dose reduction, switching to a different antipsychotic, or adding a beta-blocker or benzodiazepine.

Does Profy 15mg interact with other antidepressants?

Yes. Paroxetine and fluoxetine are strong CYP2D6 inhibitors — they significantly increase aripiprazole levels, requiring aripiprazole dose reduction by 50%. Always inform your prescriber of all medications.

Can I take Profy 15mg for depression?

Aripiprazole is approved as an adjunct treatment for MDD — added to an antidepressant when response is inadequate. It is not a standalone antidepressant and should only be used this way under specialist supervision.
